SPF EGGS


SPECIFIC PATHOGEN FREE CHICKEN (SPF) EGGS

AVS Bio provides specific pathogen free (SPF) chicken eggs for use in veterinary
and human vaccine production.

We produce SPF chicken eggs through the use of filtered-air positive-pressure
(FAPP) poultry housing, critical biosecurity measures and the careful selection
of layer flocks. Validated biological assays are utilized to confirm the SPF
status of every flock and compliance with USDA memorandum 800.65 and European
Pharmacopoeia 5.2.2 guidelines. A quality control sheet documenting this status
accompanies each product shipment. This high degree of quality control and
technical expertise is continued through product processing, packing and
shipping.

We offer three quality levels of SPF chicken eggs:

 * Premium Plus: Tested for all Research-grade agents except endogenous gs

PRIMARY CELLS


CHICKEN CELL PRODUCTS

Cell viability and virus contamination are enormous concerns when you need
pristine cell products in your drug research and manufacturing process. AVS Bio
alleviates those concerns by providing you with the industry standard for
tightly monitored, quality tissue culture cell products.

PRIMARY CHICKEN EMBRYO CELLS

We routinely prepare small and bulk production lots of chicken embryo
fibroblasts (CEF), chicken embryo kidney cells (CEKC), chicken embryo liver
cells (CELC) and chicken embryo dermal cells (CED). Custom cell preparations are
also available.

Cells are prepared fresh daily to order using our SPF chicken embryos and media
containing irradiated serum from countries with a negligible risk of bovine
spongiform encephalopathy (BSE). We offer three quality groups of cells:
standard, chicken anemia virus (CAV) negative, and CAV-negative group-specific
(GS) antigen-free cells which are C/O and not resistant to any of the Avian
Leukosis Virus subgroups.

PREPARATION OF CELLS


PROTOCOL FOR HARVESTING AND PREPARING CHICKEN EMBRYO CELLS

Cells are prepared where only tissue culture media and cell preparation are
performed and employee access is restricted. Cells are prepared under Class A
laminar flow hoods using aseptic technique throughout the harvesting process.
Batch records for all the steps in the process, including cell batch records,
documentation of the sources, media components and cell performance/viability,
are maintained. After the cells are harvested, a sample is seeded into tissue
culture flasks and maintained for 48 hours to verify cell performance. A sample
of the cells is also taken, plated onto blood agar and then incubated for 48
hours. The plates are checked after 24 and 48 hours of incubation. Checking
after 24 hours assures that bioburden is not likely present before our customers
receive the cells. If a bioburden issue occurs, customers are notified
immediately.


TISSUE CULTURE MEDIA


Basal Medium Eagle (BME) containing irradiated fetal calf serum (FCS) and/or
newborn calf serum (NBCS) sourced from negligible risk BSE countries. Sold in 1L

EGG YOLK ANTIBODIES


CHICKEN IGY ANTIBODIES


AVS Bio offers a standard three-hen, 100-egg chicken immunoglobulin Y (”IgY)
product and can customize procedures and egg yolk quantities to meet your needs.

IMMUNOGLOBULIN APPLICATIONS

Immunoglobulins have been used in applications from diagnostic assays to
therapeutics. The most widely used immunoglobulin, IgG, is derived from mammals.
Chickens, however, produce a slightly different antibody, IgY, which contains a
longer Fc region.
The greater evolutionary distance between chickens and mammals makes chickens an
attractive source of antibody production. Chickens do not recognize mammalian
proteins as “self”. Therefore these “self” proteins become more immunogenic,
leading to an increased antibody diversity in chickens compared to the mammalian
host. Therefore, if you want to increase your diagnostic assay's sensitivity to
mammalian host cell proteins (HCPs), IgY antibody is an ideal reagent to use.
There are also benefits of using IgY for prophylactic and therapeutic
non-systemic applications. IgY does not activate the mammalian complement, react
with rheumatoid factors, or bind to mammalian Fc receptors. In addition, due to
the stability of IgY at low pH conditions, it can be used to combat
gastrointestinal infections.
We have been involved in developing gastrointestinal treatments using IgY in
agriculture and respiratory treatments to prevent COVID-19.

WHY USE SPECIFIC PATHOGEN FREE (SPF) HENS FOR YOUR IGY ANTIBODY PRODUCTION?

Our hens are kept in filtered-air, positive-pressure rooms with strict
biosecurity. Hens under these conditions have a naïve immune system, which
allows them to ramp up IgY production quickly compared to commercial layers. SPF
hens are also free of pathogens as listed under United States Department of
Agriculture (USDA) and European Pharmacopeia (Ph. Eur.) guidelines leading to
less risk of contamination in your product.

CHICKEN IGY ANTIBODY PRODUCTION PROCESS

Our standard chicken IgY antibody production process includes three SPF hens per
customer supplied antigen. One hundred eggs are collected over a three-month
period. Pre-immune blood samples are tested on ELISA and Western Blot before
each bird is inoculated with antigen. Boosts and serum sampling occur every
other week until week six, when egg collection begins. Then, boosts and serum
sampling continue every two weeks, and the yolks are separated from the eggs and
frozen weekly basis until 100 yolks in total are processed. IgY antibody
extraction occurs via water dilution and pH change and is concentrated and
bottled for shipment on dry ice.

CHICKEN IMMUNIZATION STRATEGY


3R IMPACT

SPF chickens are efficient producers of immunoglobulin Y antibodies and a single
egg yolk can generate 100-200 mg of IgY; to put it into perspective, one hen can
produce the equivalent in one month quantity of antibodies in their eggs as a
goat. This fits perfectly into the principles of the 3Rs (Replacement,
Reduction, and Refinement).


HUMAN INFLUENZA  & SENDAI VIRUS

AVS Bio offers human influenza type A and B viruses, and Sendai virus (Cantell
strain), supplied as allantoic fluid and sucrose gradient-purified. We also
provide virus preparations with customer-supplied Biosafety Level 1 and 2
viruses. Viruses are grown in specific-pathogen-free (SPF) embryonated eggs and
final preparations can be delivered in crude (bulk fluids), concentrated,
fractioned, or purified forms. Click here for additional information about our

Chicken serum antibodies to Influenza A/PR/8/34 and Sendai Cantell are readily
available. These are prepared in our SPF chickens raised under strict
biosecurity and HEPA-filtered air under positive pressure (FAPP) rooms.

SENDAI VIRUS (CANTELL STRAIN)

The original virus was obtained from American Type Culture Collection (ATCC),
product code VR-907, identified as Parainfluenza 1, Sendai/Cantell strain. It
was passaged several times by inoculating the virus into the chorioallantoic sac
of 9 to 11 day-old embryonated SPF chicken eggs. Virus is supplied as sterile,
clarified allantoic fluid at a minimum titer of 2000 HA units/ml, or purified to
2 mg/ml.


AVIAN ANTIGENS & ANTISERA

AVS Bio offers a comprehensive portfolio of antisera and antigen reagents, which
are available in liquid and lyophilized aliquots for most common poultry
pathogens.


AVIAN ANTIGENS

Biological preparations of common poultry pathogens prepared in specific
pathogen free (SPF) premium eggs, cell cultures or artificial media are
available in a variety of forms, such as tissue homogenates, concentrates and
allantoic fluid. All reagents are tested for potency, verified for specificity
and can be used for applications such as agar gel immunodiffusion, HI, VN, SN,

AVIAN ANTISERA

Monospecific polyclonal antisera are produced in our CAV-negative SPF chickens.
The birds used in the production of antiserum are housed in filtered-air
positive-pressure (FAPP) barrier facilities. The birds are monitored to optimize
antibody levels and to ensure the best product possible. Our standardized
antisera are produced to obtain high levels of neutralizing activity, but also
have been used for other laboratory applications, such as immunodiffusion,
hemagglutination inhibition (HI) and immunoblotting. More than 40 tests are
performed on each antiserum, and a quality control sheet is provided that
confirms specificity.

SERUM PLATE AGGLUTINATION TEST PRINCIPLE

The serum plate agglutination (SPA) test is used to detect specific antibodies
that will bind to an antigen and cause visible “clumping” or agglutination. The
antigen is prepared from cultures of Mycoplasma gallisepticum (MG), Mycoplasma
meleagridis (MM), Mycoplasma synoviae (MS), or Salmonella pullorum and a dye is
added to improve visibility of the reactions. A prescribed amount of antigen is
placed on a solid support, such as a glass plate or mirror, keeping each drop of
antigen separate. An equal amount of test serum is placed next to the antigen
and these are then mixed together. After a short incubation, the mixture is
examined for evidence of agglutination, which appears as discrete clumps of the
stained particles with a clearer background. If no antibodies are detected, the

AVS Bio offers outsourcing of virus preparations in SPF embryonated eggs, tissue
culture and poultry. We offer pre-incubated extracted embryos and avian cell
production that can be produced in large or small quantities to meet your needs.
In addition to supporting your antigen production, our laboratories provide
progeny challenge studies and commercial poultry virus isolation. Isolated virus
can be expanded upon request. Fractional dose studies can be provided to support
your product’s technical data and marketing efforts. Custom research projects
and field safety trials can also help speed your progress to regulatory
approval.

1961

Originally founded as SPAFAS (Specific Pathogen Free Avian Supplies), we started
as an egg production and testing laboratory in Connecticut


1992

We were acquired by Charles River Laboratories and have since more than doubled
our manufacturing capacity and laboratory footprint, cultivated relationships
with customers and manufacturing partners around the world, and built new

2022
